Objectives: PRF001 is the fragmented DNA polymer extracted from the testes of salmon. The purposes were to verify anti-inflammatory effect of PRF001 in CHON-001 cells in which damage was induced by H2O2, and to observe protective effect against arthritis of PRF001 intake by the rats with arthritis induced by MIA.

Methods: In vitro, Cell survival and inflammatory markers after H2O2 treatment were investigated with different concentrations of PRF001. In vivo, osteoarthritis was induced by intraarticular injection of MIA into knee joints of rats. After consumption of PRF001(10mg/kg, 50mg/kg, and 100mg/kg) for 4 weeks, the inflammatory mediator and cytokines were investigated in articular cartilage.

Results: In vitro, cells were treated with different concentrations of PRF001 and H2O2, used in a representative inflammation model to verify protein expression, and PRF001 was an inducing substance with an anti-inflammatory effect. The level of inflammatory markers, IL-1b, TNF-a, COX-2, iNOS, and PGE2, were found to be significantly decreased. In vivo, inflammatory mediators and cytokines, such as IL-1b, Erk, NF-kB, TNF-a, COX-2, and PGE2, as well as MMP3 and MMP7, which have catabolic activity in chondrocytes, were decreased in the MIA-induced osteoarthritis rats following intake of PRF001. In histological analysis, PRF001 had a protective effect on the articular cartilage.

Conclusions: PRF001 was highly effective in reducing a degradation of chondrocytes in articular cartilage. Altogether with the results, anti-inflammatory property of PRF001 contributes to the protective effects in osteoarthritis through deregulating IL-1b, TNF-a and its subsequent signals such as Erk, NF-kB, COX-2, PGE2 and MMPs.
